(a) preparation before paraffin sectioning
1. Slicing knife. Before using the traditional slicing knife, it is necessary to check the extent of the knife edge damage under a microscope and then sharpen it. Disposable blades can now also be used, eliminating the need for sharpening.
2. Trimming the wax block: Cut off too much paraffin around the tissue. Generally, there is a paraffin edge of about 1~2mm around the tissue. Both sides must be parallel.
3. The wax block is fixed: The trimmed wax block is first fixed on a small square block or metal block prepared in advance. The fixing method is to heat the wax shovel first on the alcohol lamp, and then the bottom of the wax block is melted and melted quickly. Small square block or metal block base.
4. After the slides are wiped clean, an adhesive (protein glycerin, APES, or polylysine) is coated on the slides, and different adhesives are selected according to the staining method.
